href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-1\"><em>1<\/em><\/a>) elucidate two long-standing problems in animal evolution: the ancient function of the homeobox\u00a0<em>(Hox)<\/em>\u00a0gene cluster, which has puzzled scientists for decades (<a id=\"xref-ref-2-1\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-2\"><em>2<\/em><\/a>), and the centuries-old debate on the emergence of the segmented animal body.<\/p>\n<p id=\"p-5\"><em>Hox<\/em>\u00a0genes were first discovered in flies and mice, where they specify different body segments along the anterior-posterior (A-P) axis (<a id=\"xref-ref-3-1\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-3\"><em>3<\/em><\/a>). Although their expression often overlaps in posterior body regions, they show spatially distinct anterior expression boundaries (<a id=\"xref-ref-4-1\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-4\"><em>4<\/em><\/a>) (see the figure). Importantly, the A-P sequence of\u00a0<em>Hox<\/em>gene expression in the body matches their 3\u2032 to 5\u2032 sequential occurrence within a chromosome cluster, a principle called spatial collinearity. Moreover, the more anteriorly expressed 3\u2032\u00a0<em>Hox<\/em>\u00a0genes are often expressed earlier in development, which is called temporal collinearity. In addition, individual Hox proteins are typically active close to their anterior expression boundary, because the more 5\u2032, or posterior, proteins counteract the function of the more 39, or anterior, ones whenever both products co-exist. This is called posterior prevalence (<a id=\"xref-ref-4-2\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-4\"><em>4<\/em><\/a>).<\/p>\n<p id=\"p-6\">Making sense of these rules has been challenging. For vertebrates,\u00a0<em>Hox<\/em>\u00a0collinearity and posterior prevalence are explained with the sequential activation of these genes from within a posterior growth zone (<a id=\"xref-ref-5-1\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-5\"><em>5<\/em><\/a>). This growth zone iteratively produces mesodermal body segments, called somites, and pushes them toward the anterior. If, during this process,\u00a0<em>Hox<\/em>\u00a0genes are sequentially activated and stay on in the newly generated segments, then staggered expression of\u00a0<em>Hox<\/em>\u00a0genes arises along the A-P axis.<\/p>\n<p>What is the relevance of spatial collinearity for invertebrates? Collinear\u00a0<em>Hox<\/em>\u00a0gene activity is described for patterning ectodermal derivatives such as the nervous system in insects (<a id=\"xref-ref-3-2\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-3\"><em>3<\/em><\/a>). Few studies revealed\u00a0<em>Hox<\/em>\u00a0gene expression in mesodermal structures that resemble vertebrate somites. Moreover,\u00a0<em>Hox<\/em>\u00a0gene clusters are active in both segmented and unsegmented invertebrates such as sea urchins (6). Accordingly, He\u00a0<em>et al.<\/em>\u00a0investigated\u00a0<em>Hox<\/em>\u00a0gene function in the cnidarian\u00a0<em>Nematostella vectensis<\/em>, the starlet sea anemone. Cnidarians are our most distant relatives to possess a\u00a0<em>Hox<\/em>\u00a0cluster (see the figure). Their inner surface is folded, so that their primitive gut is subdivided into chambers, called gastric pouches. These are also continuous with the lumen of the tentacles. The cnidarian lineage diverged from ours when a cluster of only three\u00a0<em>Hox<\/em>\u00a0genes existed (<a id=\"xref-ref-8-1\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-8\"><em>8<\/em><\/a>), with one anterior (3\u2032), one middle, and one posterior (5\u2032) gene. The sea anemone has a fragmented version of the ancient three-gene\u00a0<em>Hox<\/em>\u00a0cluster, with additional, lineage-specific duplications (<a id=\"xref-ref-1-2\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-1\"><em>1<\/em><\/a>). Previous expression comparisons revealed expression of\u00a0<em>N. vectensis Hox<\/em>\u00a0genes in the developing gastric pouches, with distinct\u00a0<em>Hox<\/em>\u00a0gene expression boundaries matching the epithelial boundaries between the pouches (<a id=\"xref-ref-9-1\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-9\"><em>9<\/em><\/a>\u2013<a id=\"xref-ref-11-1\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-11\"><em>11<\/em><\/a>). These boundaries are collinear with the assumed sequence of genes in the cnidarian-bilaterian ancestor [referred to as trans-spatial collinearity (<a id=\"xref-ref-4-3\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-4\"><em>4<\/em><\/a>)].<\/p>\n<p id=\"p-10\">He\u00a0<em>et al.<\/em>\u00a0established gene ablation techniques in\u00a0<em>N. vectensis<\/em>\u00a0to investigate the function of the\u00a0<em>Hox<\/em>\u00a0genes. They uncovered a fundamental role of these gene products in the formation of the gastric pouches. In the absence of Hox activity, the epithelial folds separating these pouches are lost. Therefore, similar to vertebrates,\u00a0<em>N. vectensis Hox<\/em>\u00a0genes differentially and specifically control the generation of boundaries between repeated body parts, in conjunction with a later role in controlling the different fates of these parts (<a id=\"xref-ref-12-1\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-12\"><em>12<\/em><\/a>). He\u00a0<em>et al.<\/em>\u00a0also show that the\u00a0<em>N. vectensis Hox<\/em>\u00a0genes are expressed sequentially, in concert with the stepwise appearance of folds during polyp development. Thus,\u00a0<em>N. vectensis Hox<\/em>\u00a0genes show spatial and temporal collinearity. Additionally, the loss of each\u00a0<em>Hox<\/em>\u00a0gene abolishes the epithelial folding at the most anterior expression limit of that gene\u2014where they are not coexpressed with a more posterior\u00a0<em>Hox<\/em>\u00a0gene. In the absence of the boundary formed by the folds, the two adjacent pouches fuse. These phenotypes are consistent with the posterior prevalence principle.<\/p>\n<p id=\"p-11\">This is strong evidence that the link between\u00a0<em>Hox<\/em>\u00a0gene function and some kind of body segmentation is ancestral. But how does\u00a0<em>N. vectensis<\/em>\u00a0segmentation\u2014the sequential generation of gastric pouches by epithelial folding\u2014relate to bilaterian segmentation? The simple mode of epithelial folding observed in cnidarians has inspired morphologists for more than 150 years, who saw similarities with mesodermal segmentation in bilaterians. Bilaterian mesodermal somites often emerge as outpocketings from the gut. The enterocoel theory proposed that both cnidarian pouches and bilaterian somites are derived from such outpocketings (<a id=\"xref-ref-7-2\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-7\"><em>7<\/em><\/a>,\u00a0<a id=\"xref-ref-13-1\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-13\"><em>13<\/em><\/a>). The findings of He\u00a0<em>et al.<\/em>\u00a0support this theory.<\/p>\n<p id=\"p-12\">One possible caveat is that the sequentially emerging\u00a0<em>N. vectensis<\/em>\u00a0folds are not generated from a posterior growth zone. This might represent a secondary simplification of cnidarian development, given that in the tube anemones (Cerianthida), which represent an early diverging branch of Cnidaria, additional folds are pushed forwards from a terminal growth zone (<a id=\"xref-ref-9-2\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-9\"><em>9<\/em><\/a>). Examining the expression of\u00a0<em>Hox<\/em>\u00a0genes and of growth zone markers in these cnidarians could be especially rewarding, as it might establish a similar link between\u00a0<em>Hox<\/em>\u00a0spatial and temporal collinearity and the generation of body segments from a growth zone as is observed in vertebrates (<a id=\"xref-ref-4-4\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-4\"><em>4<\/em><\/a>,\u00a0<a id=\"xref-ref-5-2\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-5\"><em>5<\/em><\/a>). Another note of caution concerns the unsolved axial relationships between cnidarians and bilaterians (<a id=\"xref-ref-14-1\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-14\"><em>14<\/em><\/a>), which led to conflicting views about the nature of the cnidarian\u00a0<em>Hox<\/em>\u00a0axis (<a id=\"xref-ref-15-1\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-15\"><em>15<\/em><\/a>). However, the data of He\u00a0<em>et al.<\/em>\u00a0seem to firmly settle this issue.<\/p>\n<p id=\"p-13\">Twenty-five years after the revolutionary zootype concept, which considered the\u00a0<em>Hox<\/em>\u00a0genes a shared feature of all animals (<a id=\"xref-ref-2-2\" class=\"xref-bibr\" href=\"http:\/\/science.sciencemag.org\/content\/361\/6409\/1310?rss=1#ref-2\"><em>2<\/em><\/a>), we can now refine this idea and propose that the\u00a0<em>Hox<\/em>\u00a0gene cluster evolved with the emergence of segmental epithelial folds and pouches in the cnidarian-bilaterian ancestor.
